miRNAs and other class of non-coding RNAs
miRNA target sites
Using USCS browser with
TargetScan, miRNA target sites were predicted on 3'UTR of the MLL gene.
Other class of Non-coding RNAs
NONCODE online blast using MLL mRNA sequence indicated that the MLL mRNA sequence contains a class of non-coding RNA,
piwi-interacting RNA (piRNA).
Protein/Domains
1. Entrez provided mRNA sequence, which is validated, and was used for further protein analysis.

UniGene identified sequence similarities and the protein sequences were compared with others (with known function) using
ClustalW.

InterPro database was used to characterise domains within the protein coded by MLL (
IPR015722 MLL Transcription Factor). The results showed some of the regions identified by protein sequence alignment.

Zinc finger protein HRX", which possibly acts as a transcriptional regulatory factor and promotes
PPP1R15A-induced apoptosis.
Zinc finger protein is a DNA-binding protein domain comprised of Zinc fingers. They occur in nature as the part of transcription factors conferring DNA sequence specificity as the DNA-binding domain.
Regulation of transcription requires interactions between transcriptional activators and transcriptional co-activator CREB binding protein (CBP). The figure shows the interactin of
Zinc finger protein HRX with Myb proto-oncogene and Crebbp protein.
SET domain is an 130- to 140-amino acid, evolutionary well conserved sequence motif that was initially characterized in the Drosophila proteins Su(var)3-9, Enhancer-of-zeste and Trithorax. In addition to these chromosomal proteins modulating gene activities and/or chromatin structure, the SET domain is found in proteins of diverse functions ranging from yeast to mammals, but also including some bacteria and viruses. The SET domains of mammalian
SUV39H1 and 2 and fission yeast clr4 have been shown to be necessary for the methylation of lysine-9 in the histone H3 N-terminus.
Protein Interaction
String-EMBL was used to seaerch for protein interaction for Zinc finger protein HRX ("MLL" in the figure).
